Collect about five CC. of spinal fluid in a sterile test tube containing a trace of Potassium Oxalate to prevent coagulation. The specimen should contain no visible trace of blood. Fill the capillary of a 1:10 pipette to the graduation with the diluting fluid and then draw the spinal fluid to the graduation above the bulb. Shake the pipette and charge the chamber as with blood corpuscle counting technique. Allow several minutes for settling. The chambers are ruled with the Fuchs-Rosenthal pattern. This consists of 16 one square millimeter areas orientated by triple lines, and each such area sub-divided into 16 squares. It is generally recommended to count 16 one square millimeter areas preferably 8 in each chamber.

1) Weigh out 2 grams of feces

2) Pass feces through a sieve into a dish containing 60 ml of ZnSO4 or saturated salt solution.

3) Stirring vigorously (transfer solution to a flask to prevent spillage), take a sample with a pipette or medicine dropper and load one of the chamber of the slide. Repeat and fill other chamber.

4) Wait 30 seconds for the eggs to float to the upper surface of the counting chamber.Two focal planes exist, with the eggs an air bubbles in the upper plane. Focus on air bubbles, then count the total number of eggs under both of the grids. Do not count eggs outside of the grid.

5) Multiply the total number of eggs in the two chambers by 100, this is the eggs per gram(EPG).

To clean the counting chamber: After completing the count, remove the cover glass and clean the counting chamber with water or a mild cleaning solution (10% solution of bleach). Dry the counting chamber with a soft cloth or wipe, or rinse with acetone.

The Speirs-Levy Eosinophil counting slide is designed primarily for doing direct eosinphil counts in blood, but is readily adaptable for performing cell counts in spinal fluids or other media in which cells are suspended. It is especially useful in clinics and laboratories where large number of blood cell determinations must be performed quickly.

The Speirs-Levy slide has the following characteristics which facilitate loading and counting:

1. There are 4 chambers per slide

2. Each chamber consists of ten 1 x 1mm squares which are subdivided into 16 smaller areas.

3. The depth of each chamber is 0.2 mm.

4. The chambers are spaced on the left hand side of the slide for ease of handling and to prevent accidental reversing of the slide.

5. An identification number is engraved on the right hand side of each chamber and will appear on the left hand side when viewed through the microscope. This number helps focusing and may be used to determine the beginning and end of a chamber count.

6. The outer edge of each chamber is inclined 7 degrees to aid in proper charging of the chamber and to act as a reservoir to replace any fluid that might evaporate before the slide is counted.

The Counting Chamber is of all glass construction. In the center of the slide is a rectangle 15 x 20 mm, which is flanked by shoulders on each side 0.1mm higher. The cover glass is supported on these shoulders and leaves a depth of 0.1mm between the underside of the cover glass and the rectangle. The rectangle and the cover glass have optically plane surfaces. To facilitate calibration of the microscope, the rectangle has two engraved parallel lines spaced 1.382mm apart.

An accessory eyepiece micrometer is available which is ruled into squares, each of which is equal to 16 of the diameter of the eyepiece diaphragm opening. For mold counting, the microscope must give an area of 1.5 sq. mm.(circle 1.382 mm in diameter) at magnification of 90-125.
