Gardenoside is one of the most active gardenia ingredients, but if we need to purchase gardenoside, how do we determine the amount of gardenoside?
currently, the methods used to determine gardenoside content are high performance liquid chromatography (hplc), thin layer scanning (tlc) and thin layer - uv spectrophotometry. When using hplc, gardenoside's hydrophilicity is used, and the mobile phase typically uses phosphate aqueous solvents and polar solvents, compared to the standard gardenoside chromatograms. Thin-layer scanning is used because gardenoside has fluorescent absorption spots at 254m in the uv region. Using this property to locate under fluorescent lamps, uv scanning is simple, stable and reproducibility. For the determination of gardenoside by tlc-uv spectrophotometry, a suitable silica gel thin layer plate and an unfolding agent are required. The unfolding agent can use chloroform: methanol =3:1, and the wavelength obtained is 237 nm. Gardenoside sample solution is required to be prepared for determination.
